RESUMEN

Desmodium styracifolium is a medicinal plant from the Desmodieae tribe, also known as Grona styracifolia. Its role in the treatment of urolithiasis, urinary infections, and cholelithiasis has previously been widely documented. The complete chloroplast genome sequence of D. Styracifolium is 149,155 bp in length with a GC content of 35.2%. It is composed of a large single copy (LSC) of 82,476 bp and a small single copy (SSC) of 18,439 bp, which are separated by a pair of inverted repeats (IR) of 24,120 bp each and has 128 genes. We performed a comparative analysis of the D. styracifolium cpDNA with the genome of previously investigated members of the Sesamoidea tribe and on the outgroup from its Phaseolinae sister tribe. The size of all seven cpDNAs ranged from 148,814 bp to 151,217 bp in length due to the contraction and expansion of the IR/SC boundaries. The gene orientation of the SSC region in D. styracifolium was inverted in comparison with the other six studied species. Furthermore, the sequence divergence of the IR regions was significantly lower than that of the LSC and the SSC, and five highly divergent regions, trnL-UAA-trnT-UGU, psaJ-ycf4, psbE-petL, rpl36-rps8, and rpl32-trnL-UGA, were identified that could be used as valuable molecular markers in future taxonomic studies and phylogenetic constructions.

Polyhydroxyalkanoates (PHAs) including poly-3-hydroxybutyrate (P3HB) as secondary metabolisms were in vitro produced by the edible basidiomycetous mushroom Astraeus odoratus during its growth on malt agar extract. Various carbon and nitrogen sources containing cellulose, glucose, glycerol, rice straw powder, soybean meal and peptone were investigated for the growth of basidiomycetous mushrooms. During cultivation, the A. odoratus culture exudated the considerably extracellular fluid up to approx. 2.3 ml on 2% malt extract agar plate within 7 days. The chemical compounds of the exudated fluid were further investigated by Fourier-transform infrared spectroscopy (FTIR) and gas chromatography/mass spectrometry (GC/MS); and its morphology of the lyophilized sample was observed by scanning electron microscope (SEM). FTIR results showed the characteristic bands of OH at 3445 cm-1, CH/CH2/symmetric CH3 (stretch) at 2923 and 2852 cm-1, C=O at 1730 cm-1, asymmetric CH3 (bend) at 1454 and 1414 cm-1, C-O of COO- at 1396 cm-1 and C-O-C at 1223, 1160, 1116, 1058 and 1019 cm-1 which were similar to the absorptive characteristics of P3HB. Methyl ester derivatives of GC/MS results identified 7 compounds including: 3-hydroxybutanoic (monomer of PHB), aminobenzoic, salicylic, hexadecenoic, octadecadienoic, octadecenoic and octadecanoic acids. SEM images revealed a fibriform and porous materials. Hence, the occurrence of PHAs was first described in a basidiomycetous mushroom A. odoratus. Thus, PHAs could be found not only in bacteria and but also in basidiomycetous mushroom, which can be promising target for bioplastics and green environmental studies.

RESUMEN

The complete chloroplast genome (cpDNA) Viburnum odoratissimumis sequenced and assembled from the whole genome data. The cpDNA of V. odoratissimum is 158,744bp in lengthwith the overall GC content of 38.1%. It consists of a pair of inverted repeats (IRa and IRb, 26,494bp) which separate a large single copy (LCS, 87,348 bp) and small single copy (SSC,18,408 bp). The complete chloroplast genome contains 129 genes, including 84 protein-coding genes, 8 rRNA genes and 37 tRNA genes. A phylogenetic tree of DNA sequences of barcoding regions, including rbcL, matK, psbA-trnHfrom 8 species of the Genus Viburnum shows that V. odoratissimum is closely related to V. furcatum and V. burejaeticum.

Lentinus arcularius (=Polyporus arcularius) is a medicinal polypore mushroom recorded in Vietnam based on morphological observation. In this study, three L. arcularius isolates were identified using molecular and morphological methods. Furthermore, the antimicrobial and antioxidant activities of their intracellular and/or extracellular metabolites were investigated. The ethyl acetate and n-butanol fractions were the most active extracts against Gram-positive and Gram-negative bacteria (Escherichia coli VTCC 12014, Pseudomonas aeruginosa VTCC 12273, and Staphylococcus aureus VTCC 10658) and fungi (Candida albicans VTCC 20568, Saccharomyces cerevisiae VTCC 20146, and Aspergillus niger VTCC 30001). These fractions also showed the highest antioxidant activity by α,α-diphenyl-ß-picryl-hydrazyl as a free-radical agent or the highest percentage at 75-100% inhibition. These results suggest that L. arcularius might be a potential medicinal mushroom with antimicrobial and antioxidant effects.

Sweet marjoram (Origanum majorana) is an aromatic herb in the Lamiaceae family. This study aims to report the complete chloroplast nucleotide sequence of marjoram and the phylogenetic relationship with other Lamiaceae species. The total length of this plastome is 151,841 bp, containing a pair of inverted repeat regions (25,558 bp), separated by a large single copy region (83,035 bp) and a small single copy (17,690 bp). The genome encodes 132 genes, including 86 protein-coding genes, 36 tRNA genes, and eight rRNA genes. Phylogenetic tree analysis indicated that O. majorana is the most closely related to Origanum vulgare.

BACKGROUND: The overall goal of the Supporting Adolescent Adherence in Vietnam (SAAV) study is to improve understanding of an adherence feedback mHealth intervention designed to help adolescents living with HIV (ALHIV) maintain high adherence to antiretroviral therapy (ART), critical to effective treatment. Specifically, we aim to: (1) conduct formative research with Vietnamese ALHIV and their caregivers to better understand adherence challenges and refine the personalized mHealth intervention package; and (2) assess the feasibility, acceptability, and efficacy of the intervention to improve ART adherence by implementing a randomized controlled trial (RCT). METHODS: The study will utilize mixed methods. The formative phase will include 40 in-depth interviews (IDIs) with 20 adolescent (12-17 years)/caregiver dyads and eight focus group discussions with adolescents, caregivers, and clinicians at the National Hospital for Pediatrics (NHP) in Hanoi, Vietnam. We will also conduct 20 IDIs with older adolescents (18-21 years) who have transitioned to adult care at outpatient clinics in Hanoi. We will then implement a seven-month RCT at NHP. We will recruit 80 adolescents on ART, monitor their adherence for one month to establish baseline adherence using a wireless pill container (WPC), and then randomize participants to intervention versus control within optimal (≥ 95% on-time doses) versus suboptimal (< 95% on-time doses) baseline adherence strata. Intervention participants will receive a reminder of their choice (cellphone text message/call or bottle-based flash/alarm), triggered when they miss a dose, and engage in monthly counseling informed by their adherence data. Comparison participants will receive usual care and offer of counseling at routine monthly clinic visits. After six months, we will compare ART adherence, CD4 count, and HIV viral suppression between arms, in addition to acceptability and feasibility of the intervention. DISCUSSION: Findings will contribute valuable information on perceived barriers and facilitators affecting adolescents' ART adherence, mHealth approaches as adherence support tools for ALHIV, and factors affecting adolescents' ART adherence. This information will be useful to researchers, medical personnel, and policy-makers as they develop and implement adherence programs for ALHIV, with potential relevance to other chronic diseases during transition from adolescent to adult care. A core-shell cerium oxide nanorod@polypyrrole (CeO2-NR@Ppy) nanocomposite-based electrochemical DNA biosensor was studied for Salmonella detection. The core-shell CeO2-NR@Ppy nanocomposite was prepared by in situ chemical oxidative polymerization of pyrrole monomer on CeO2-NRs, which provided a suitable platform for electrochemical DNA biosensor fabrication. The immobilization of ss-DNA sequences onto nanocomposite-coated microelectrode was performed via covalent attachment method. DNA biosensor electrochemical responses were studied by cyclic voltammetry and electrochemical impedance spectroscopy with [Fe (CN)6]3-/4- as redox probe. Under optimal conditions, DNA biosensor response showed good linearity in the range of 0.01-0.4 nM with sensitivity of 593.7â€¯Ω·nM-1·cm-2. The low limit of detection and limit of quantification for the DNA biosensor were 0.084 and 0.28 nM, respectively. The proposed DNA biosensor also showed good results when used in detecting actual Salmonella samples.

Fusarium proliferatum NBRC109045 is a filamentous fungus isolated from Vietnamese forest due to high production of ß-glucosidases. Production of the enzyme was studied on varied carbon source based mediums. The highest activity was obtained in medium containing 1% corn stover + 1% wheat bran (3.31 ± 0.14 U/ml). It is interesting to note that glucose (0.69 ± 0.02 U/ml) gave higher activity and just followed by cellobiose among the di- and mono-saccharides, which is generally regarded as a universal repressor of hydrolases. We improved the zymogram method to prove that in response to various carbon sources, F. proliferatum could express various ß-glucosidases. One of the ß-glucosidases produced by F. proliferatum growing in corn stover + wheat bran based medium was partially purified and proved to have high catalytic ability.
